Description
This is a composite part of an autodisplay technology translocating a fluorescent protein (mCherry). This part includes an T7 promoter with a lacI operator as well as an RBS(AGGA). The fluorescent protein will cleave post-translationally. This part can be used to quantify the surface density and activity of autodisplay technology. PET is a beta-barrel autodisplay technology native to E.coli. mCherry is a derivative of RFP.
Notes
To comply with BioBrick standard 10, Phe-76 was re-optimized to remove a EcoRI site within the part. A T7 promoter with a lacI operator was added with the RBS (AGGA) to the PET_mCherry. A standard assembly 10 prefix and suffix was then added.

Microbial Cell Factories. 2012. The sequence was assembled from synthesized 500bp blocks through IDTdna "Gblock" synthesis.
