Description
Our team use this part as a final vector for TALE(TranscriptionActivator???Like Effectors, TALE) assembly via Golden Gate Assembly. When we insert the TALE repeats into this vector, the reporter gene LacZ will be knocked out by the restriction enzyme Esp3I. Then we can use blue-white screening to select the right white colonies by add 40μL of 20mg/mL X-gal +40μL of 0.1M IPTG into the plate.Notes
The N-terminal of TALE contains the restriction sites of PstI. Therefore, users should note if they want to use PstI to digest this plasmid.Source
