Description

This part is for specific transport of CsgA(BBa_K1361999) and CsgB(BBa_K1361997) into extracellular matrix.
CsgE is a small periplasmic protein, is proposed to direct CsgA to the CsgG secretion complex and helps mediate secretion specificity via an N-terminal sequence in the mature CsgA.
CsgF is a membrane-associated protein, required for CsgB surface exposure, and is crucial for efficient CsgA polymerization.
CsgG is a canal protein at outer membrane that is required for secretion of CsgA and CsgB into the extracellular milieu.

Notes

The CsgEFG was PCRed as a whole, despite there may be some regulation sequence between them. The EcoRI loci within CsgA sequence has been replaced from GAATTC to GAATTT.

Source

PCR on genomic DNA from E coli. DH5alpha strain. And site-directed mutation by overlap PCR primers.
