Description
This is the final construct from one of the projects of Caltech 2008 iGEM team. In response to being induced with acylhomoserine lactone, the construct should cause the E. coli to produce pyruvate oxidase (SpxB) while simulataneously shutting down production of a catalase, KatG. KatG is cleared rapidly from the cell due to an LAA degradation tag, allowing hydrogen peroxide produced from the pyruvate oxidase to build to bactericidal concentrations. Preliminary data suggests pyruvate oxidase can produce 0.8 mM hydrogen peroxide in the media supernatant.
