Description

This consists of melittin fused to GST (Glutathione S-Tranferase) with a TEV cleavage site. Melittin is a toxin which causes cell death. GST-melittin is an inactive form of melittin, which ensures that melittin can be produced in a cell without killing the host cell. To activate melittin, TEV protease is added to cleave melittin from GST.

Notes

Thrombin was the initial cleavage site, however we decided to use TEV because it was more efficient. As a result, we have both thrombin and TEV cleavage sites.
