Description

We constructed and improved standard BioBrick Parts about cyanide detoxification.This part conteins CynR,CynT,CynS three genes.The CynT and CynS belong to cynTSX operon. The transcriptional activation of the cynTSX polycistronic message is under the positive control of the CynR protein and cyanide.
The highlight is that the cynTSX operon enables Escherichia coli K-12 to degrade and use cyanide as a sole nitrogen source. And the most important point is the promoter of this operon is positively regulated by cyanide and the CynR protein, which means this part can play a role only in the exist of cyanide.

Notes

The CynR gene is located upstream of the cynTSX operon and is transcribed in the opposite direction of the cyn operon. CynR negatively regulates its own synthesis independently of cyanide. What???s more,the cynR promoter was found to overlap the cynTSX promoter on the opposite strand.
Moreover furthermore,cyanate degradation requires carbonic anhydrase encoded by cynT and cyanide hydrolase encoded by cynS.CynS catalyzes the bicarbonate-dependent hydrolysis of cyanide into ammonia and two molecules of carbon dioxide. CynT recycles the carbon dioxide into bicarbonate.The third gene of the cyn operon, cynX, encodes a hydrophobic protein of undetermined function.So we didn???t choose it.
