Description

It is a TetR repressible promoter coupled with fadL, fadD genes and strong RBS. It is inducible by atc to generate fadD and fadL intermembrane proteins. The long-chain fatty acid (LCFA) transporter encoded by fadL will transport LCFA through the outer membrane. The acyl-CoA synthase encoded by fadD gene will further activated LCFA into long chain acyl-CoA and transport them through the inner membrane to the cell.

Overexpressing fadD and fadL will render the E.coli strain a higher LCFA intake and extra source of LCFA for its metabolism or other biosynthetic purpose.

This protein generator was used by CityU_HK 2014 team to increase the LCFA uptake for alpha-Linolenic acid production in E.coli.

Notes

The fadD and fadL genes are obtain from PCR amplification but the sequences are the same as the BBa_K861500 and BBa_K861010. This construct is relying on the built in terminator of the biobrick plasmid.

It is constructed from standard registry RBS (BBa_B0034, BBa_B0030) and promoter (BBa_R0040). It is coupled with fadD and fadL genes amplified from the genome of E.coli K-12 strains.
