Sequences BBa_K1509002_sequence (Version 1)

Description

It is responsible for the flocculent ability of Bacillus sp. F2 and has been converted to E.coli JM109. It has been proved that strain of E.coli positive clone FC2 could express flocculent activity, with the flocculent efficiency of 90%, thus can be a good choice in environmental applications. The 1071bp gene consists of 29%T, 18%C, 25%G and 29%A.

Notes

It was attained from Bacillus sp. F2 by using PCR with the total genomic DNA of F2 as template. The two synthesized oligonucleotide primers were shown as below:
P1: 5???-AGTCTAGATGTAAGCTCTCTTCCGG-3???
P2: 5???-CTCTCGAGCAATAAGGACGCAATG-3???
And the PCR runs under the settings as follows:
Temperature Number of cycle
96℃; 15min 1
94℃; 40sec 30
50℃; 60sec
72℃; 4min
72℃; 7min 1
Then it was spliced with the DNA sequence for peptide CDS 7 with a synthetic constitutive promoters CP25.

Source

It???s origin is one strain named as Bacillus sp. F2, whose flocculent ability can reach 84%.
