Description
HFBI is a modle of class I of hydrophobins, which are naturally produced by filamentous fungi. We use serine, which is similar with cysteine in structure, to replace all of its conserved eight-cysteine residues and get the mutant of HFBI. Hydrophobins are amphipathic, which means they have the hydrophobic part and hydropholic part at the same time. HFBI is a kind of hydrophobins and it has magic properties. We combine GFP with HFBI by a linker and make them become a fusion protein. This fusion protein could be used in aqueous two-phase systems to make itself seperated frome mixture. Meanwile, hydrophobin has a property that it can generate a membrane on the surface of solid automatically, which is called self-assembly. We can get a fluorescent protein' membrane by it.
Notes
When amplify the HFBI and GFP with PCR, temperature is important.
Source
GFP comes frome iGEM's standard part. We construct the mutant of HFBI ourselves, and the original HFBI comes from Trichoderma reesei.
