Description
Replication of tour bioBrick can via integration in B. subtilis locus. One of these loci is the ThrC locus. This backbone is based on the plasmids pdg1664 and K823023. The AmyE loci from K823023 were replaced with the ThrC loci from pdg1664. Along with this exchange, the chloramphenicol resistance gene was replaced with erythromycin resistant gene (wich was also found in the pdg1664). This biobrick can be replicated both in E. coli and in B. subtilis, where it intergrates into the genome.
Notes
pdg1664 contained an EcoRI and a XbaI restriction site. However these sites were situated around a locus. An option was to introduce the prefix, suffix and remove the other restriction site. Here is chosen to copy the loci and erythormycin resistance gene and use these to replace the AmyE intergration loci and chloremphencicol resistance gene from Bba_K823023.
