Description
Bacillus subtilis RibD gene for riboflavin production. RibD code for the Pyrimidine reductase/deaminase.
Coding sequence has been codon optimized for Lactobacillus plantarum. The gene is promoted by the synthetic promoter p25 from 'A synthetic promoter library for constitutive gene expression in Lactobacillus plantarum', Microbiology, 2006.
The gene is terminated by the Tldh terminator (Spath et al. Microbial Cell Factories 2012, 11:141)
The RBS and the spacing between the RBS and the coding sequence are optimized for Lactobacillus plantarum (Tauer et al. Microbial Cell Factories 2014, 13:150).
The expressed protein catalyses two steps in the riboflavin pathway.
Notes
RibD is not considered as a bottleneck in the riboflavin pathway. A medium promoter is so expressing it.
Source
Coding sequence comes from Bacillus subtilis genome sequence. It has been codon optimized for Lactobaciilus plantarum on IDT website.
p25 is a synthetic constitutive promoter designed in 'A synthetic promoter library for constitutive gene expression in Lactobacillus plantarum', Microbiology, 2006.
The RBS informations are coming from Tauer et al. Microbial Cell Factories 2014, 13:150.
Tldh terminator information are documented in Spath et al. Microbial Cell Factories 2012, 11:141.
