Description
The part codes for a synthetic luciferase and a NADPH-flavin oxidoreductase, joined by a 2A peptide sequence. Genes used are codon optimized for expression in yeast. When provided with exogenous substrate or coexpressed with the lux system fatty acid reductase complex, the part allows for light production. Luciferase catalyzes the conversion of substrate for light production, while the oxidoreductase regenerates FMNH2 for use by luciferase. The 2A peptide allows for stoichiometric expression of the two proteins.
Notes
The sequence was too long to be directly synthesized, so we had to synthesize it in fragments that were joined through Gibson Assembly.

The luciferase used is a synthetic construct, modified from Photorhabdus Luminescens luciferase and engineered for enhanced activity and thermostability. The NADPH-flavin oxidoreductase used was isolated from Vibrio Harveyi.
