Description
It encodes C-terminal of luciferase. When it meets N-terminal of luciferase, these two parts can become a complete luciferase. We cloned this gene to an expression plasmid pET30a(+) and add IPTG until its final concentration is 1mmol/L, incubate for 4-6h(37??C, 250rpm) to induce the expression of N-Luciferase. We use it as a part of reporter gene to detect whether it has taxol or its analogues.
Notes
To get this part, we designed primers according to the gene sequence and then cloned it to our expression vector.Because this gene doesn't have a initiation codon, so we designed a initiation codon in its forward primer.
Source
We get this gene from pCambia1300-C-Luciferase plasmid.
