Description
This parts is composed of a random sequence of 100 base pairs follow upstream and downstream by a BsaI restriction site. The fusion sites for both ends are part of the standard prefix and the suffix. The part has the advantage to be clone into a RFP plasmid with RFC 10 and then used as the vector for the Golden Gate reaction.
Notes
We had a random sequence into the BsaI cut sites to make a sequence easy to detect after a PCR reaction and leading the enzyme digestion. For further design a good option is to change the random sequence with a RFP o GFP device, using J23101 for example, to have a selection in the transformation.
