Description
This is DNA sequence coding for S. cerevisiae guanine nucleotide-binding protein subunit alpha (GPA2) with 5 amino acids at C terminus replaced by corresponding part of human G(s) alpha-subunit (alpha-S1). The strategy of making yeast???human chimeric G-protein, in which the carboxyl-terminal 5 amino acid residues of yeast G alpha subunit are replaced by the equivalent residues from human Gs proteins, has been shown to improve signal transmission in GPCR engineered yeast according to recent literature.
Notes
The endogenous G proteins may not be able to couple with the mammalian GPCRs efficiently, so we do the modifications at C terminal residues to yield better results.
