Description
This E. coli strain can be used to propagate plasmids with R6K conditional replication origin (
BBa_J61001,
BBa_K300998), such as
BBa_K300000 integrative base vector after the excision of its insert (
BBa_I52002).
BW23474 propagates R6K origin at a copy number comparable to high copy plasmids (~250 plasmids per cell) [1].
Genotype:
F-, Δ(argF-lac)169, ΔuidA4::pir-116, recA1, rpoS396(Am)?, endA9(del-ins)::FRT, rph-1, hsdR514, rob-1, creC510
Notes
This is not the only strain able to propagate R6K at high copy, but it has been mentioned in several papers and it is easily retrievable from the CGSC.
