Description

hsa-miR-122 binding site pattern, consisting of 4 binding sites (BS). The BS are seperated by sequences of 30 base pairs, which are innert in terms of miRNA binding. Additionally, half of the separation sequence is either in front or the end of the whole pattern, to be unconnected to standard restriction sites.

Mutations showed up for BS 1 and 2, where BS 3 and 4 are perfectly matching:
1st: CAAACACCATTGTCACACTC-A
perfect: CAAACACCATTGTCACACTCCA

2nd: CAAACACCATTGTCA-ACTCCA
perfect: CAAACACCATTGTCACACTCCA

Notes

The spacer sequences, present between the miRNA binding sites, should have the lowest compatibility to known miRNAs, to minimize unspecific sequence recognition.
