Sequences BBa_K414001_sequence (Version 1)

Description

In order for Green Fluorescent Protein to function properly in plants, a plant-specific ribosome binding site (Kozak sequence)must be added to the 5'region just upstream from the start of translation. The Kozak sequence is AAA AAA AAA ACA and is directly adjacent to the ATG start codon in GFP.

Notes

We chose a strong plant Kozak sequence that can function in a broad range of dicot species, including tobacco and Arabidopsis.

Source

The Kozak sequence was directly engineered into the prefix primer and used to PCR out the GFP gene from E0040.
