Description
This part encodes a fusion between the Delta-3 ligand and the monomeric mCherry red fluorescent protein. Additionally, the protein is succeeded by four copies of the FF4 intronic miRNA binding sequence, allowing for miRNA-mediated repression of this part's translation.
Figures 1 and 2 below show fluorescent output from the Delta-mCherry protein, while Figures 3 and 4 below show transactivation of the Notch-Gal4-ESN receptor by the Delta-mCherry protein.
Notes
Due to the high secondary structure of FF4 miRNA, this part was assembled with standard restriction cloning methods, specifically using enzymes AscI and BamHI.
