Description
This DNA sequence codes for the binary enzyme CrtYB. CrtY (Lycopene Beta-cyclase) converts lycopene into beta-carotene and serves as the last step in beta-carotene biosynthesis in Saccharomyces cerevisiae. CrtB is acts further upstream of CrtY in the pathway and is called Phytoene Synthase. CrtB converts geranylgeranyl diphosphate to phytoene.
Notes
While designing the sequence, we made sure that the cut sites for restriction enzymes avr2 and spe1 did not occur anywhere within the sequence.
Source
The sequence of this part was originally taken from the genome of Xanthophyllomyces dendrorhous. This sequence can be found in the NCBI database. We then codon optimized this sequence for Saccharomyces cerevisiae using an online tool found here: http://www.encorbio.com/protocols/Codon.htm
