Description

Screening techniques available presently are all negatively related. The disadvantages of antibiotics are:

1. Principle of Negative Selection
2. Transformants carry a small but unnecessary risk of horizontal gene transfer to environmental microbes
3. Metabolic burden on the cells that significantly reduces the production of the desired protein

Therefore, a novel selectable marker is needed which confers metabolic ???advantage??? to the transformants.

This plasmid carries the gene proteorhodopsin.

The reading frame had to be maintained.
