Description

E.coli transformed with this construct will produce a superfolder Green fluorescent Protein (sfGFP) fused variant of reflectin A1 under the control of pBAD, an arabinose inducible protein. There is currently (at the time of writing) no good assay for reflectin, in vivo. This construct allows reflectin to be localised within the cell. This part has been used to view reflectin containing inclusion bodies in E.coli and the sfGFP retained its fluorescence despite the fusion.

Notes

We deliberately arranged the sfGFP part of the fusion on the C terminus of the protein as translation is known to proceed from N terminus to C terminus. This means that any sfGFP visualised in vivo must have been produced with reflectin A1 synthesised first, then the sfGFP component.

This composite part was built using gibson assembly to add Reflectin A1 between pBAD and sfGFP on an existing pBAD-sfGFP construct.
