Description
This is a demo wetlab project performed by the 2011 NYC iGEM Software team to introduce them to basic practices of molecular biology, and to explore the steps and tools involved in putting together a simple genetic device.
E.Coli recA promoter which had been colony PCR'd out of JM109 strain with RFC23 overhangs was combined with a ribosome binding site and mRFP1 E1010 coding device.
This device is designed to produce RFP in presence of any external stimuli that can trigger the recA promoter. For purposes of NYC iGEM Software and our sister team NYC iGEM Wetware we focused on the device's ability to act as an ionizing radiation sensor.
Sequence and Features
Notes
Automated biobricking programs coupled with ability to look up genbank and other web based DNA databases speed up the ease and speed of compositing simple systems significantly.
Source
recA promoter was lifted via colony PCR from JM 109 strain of E.Coli with RFC 23 overhangs. RBS and RFP generator are existing biobricks from the registry. (B0034 and mRFP1 E1010)
