Description
Truncated Spot42 stands for Spot42 scaffold without repression stem loop,remaining Hfq binding site and endogenous terminator.We assembly other targets(expect galK) for original spot42(like nanC,which has a higher affinity than galK) by ClaI/SalI digestion and ligation,to make a buffer RNA for galK::GFP mRNA.[J23106,P0412]is a LacI generator, which represses lac promotor and can be induced by IPTG.
IPTG concentration gradient results in different sRNA Spot42 levels in cells
ClaI site and SalI site are well-selected restriction sites,they are used to assembly the artificial region to the 5??? end of Spot42,are compatible for all of the Ampr and Chlr vectors(no unexpected restction sites),whereas all the Kanar vectors will be restricted into fragments when treated with SalI/ClaI.
