Description
This part contains an engineered theophylline riboswitch (RS) (BBa K784005) and T7 RNA polymerase (BBa I2032). The riboswitch contains a RBS and it will be exposed and starts translating the T7 RNAP only after inserting of theophylline. The RBS and the start codon of the gene are part of the riboswitch sequence and are responsible for its structure, so they are not to be changed. This part was constructed using fusion PCR, so the RS and the T7 RNAP are adjacent to one another without any restriction site between them. There is an additional stop codon at the end of the gene.Notes
