Description
This tRNA terminator is based around the downstream sequence of the E. coli Ala(GCC) tRNA gene. It contains the region of 3-6 T???s that is characteristic of tRNA terminators and an additional 10 bp of native sequence. This additional region of native sequence is to ensure RNA termination and to act as a small spacer region between tRNA genes.
It is important to use tRNA specific terminators when expressing tRNA genes, as normal rho-dependent and rho-independent terminators rely on additional RNA sequences after where the gene actually finishes to terminate, and when extra sequences are included on tRNA molecules, their function can be severely affected.
