Description
Negative control for TAL repressor specificity. TAL repressor 1 expression under the control of a pBAD promoter and a theophylline riboswitch inducible by arabinose and theophylline. The TAL repressor should not be able to bind to the TBS2, since it is not its specific binding site, TBS1 (BBa_K1212004), thus GFP should be expressed if pTet is induced by aTc.
Notes
There cannot be a scar between the end of the riboswitch and the start codon of the TAL repressor.
Source
The pTet,RBS, GFP, Terminator and pBAD are parts from the registry. The riboswitch is from the Gallivan Lab's paper Synthetic riboswitches that induce gene expression in diverse bacterial species. by Topp et al. in Appl Environ Microbiol. The TAL repressor and TBS are from the Segal Lab at the UC Davis Genome Center. See their paper Quantitative Analysis of TALE-DNA Interactions Suggests Polarity Effects. by Meckler et al. submitted to Nucleic Acids Res.
