Description
This is a dual inducible expression system composed of an RFP color variant under the control of the Lac promotor (pLac) and a GFP color variant under the control of the Xyl promotor (pXyl). The addition of IPTG selectively promotes expression of RFP while the addition of xylose promotes the expression of GFP. The Xyl repressor (XylR), which binds pXyl and represses expression expression of genes under the control of that promoter, is also included in the part in order to repress GFP expression in the absence of xylose.
Notes
Because repression of pLac requires the presence of the lac repressor (LacI), bacterial strains that do not express a functional LacI (such as dh5alpha) exhibit high levels of RFP expression even in the absence of IPTG. Therefore, to achieve inducible expression of the RFP, the part must be expressed in E. coli strains that express a functional form of LacI (e.g., JM109 or BL21).
