Description
This part deletes the pilT1 ATPase gene in Synechocystis sp. PCC 6803. PilT1 normally allows for pilus retraction.
Notes
The part is designed to contained regions homologous to the pilT1 gene. As such we had to ensure that the homologous region was of sufficient length to allow for recombination. The part contains two regions homologous to pilT1: the first region is homologous to the first 200 bases of pilT1, the second region to the last 200 bases of the pilT1 gene. The centre of the part is a kanamycin resistance gene, which becomes inserted in the middle of the chromosomal pilT1 gene during recombination.
