Description
This part inserts the pilA1 gene into Synechocystis sp. PCC 6803. The part contains the pilA1 gene and the upstream Pcpc560 super-strong promoter.
PilA1 is the main pilus subunit.
Notes
The part is designed to contained regions homologous to the pilA1 gene. As such we had to ensure that the homologous region was of sufficient length to allow for recombination. The part contains two regions homologous to pilA1: the first region is homologous to the first 200 bases of pilA1, the second region to the last 200 bases of the pilT1 gene. The centre of the part contains the Pcpc560 super-strong promoter, the PilA1 gene and a kanamycin resistance gene, which become inserted in the middle of the chromosomal pilT1 gene during recombination.
