Description
Part BBa_K1584026 is a reporter part that releases green fluorescent light. It produces two proteins, first LuxR and GFP. LuxR is constantly produced with constitutive promoter J61100. When AHL is present, it binds to LuxR. The resulting compound binds to the LuxRP promoter, producing green fluorescent proteins.
It includes Parts J61100, a ribosome binding site with a constitutive promoter from Part J23100 inputted, to promote the production of LuxR, C0062. The terminator behind LuxR was from Part K83017. Part J37032 was used as a GFP DNA segment controlled by the LuxRP promoter, to produce GFP.
Notes
In order to properly produce the LuxR protein, we put a terminator behind LuxR.
We also had two XbaI sites at J61100, so when we tried to cut using EcoRI and XbaI, there was a chance that the constitutive promoter could be eliminated, which we had to consider.
