Description

pxylS-E1_5'UTR construct assembly was performed based on XylS promoter (Bba_K1741009). XylS consist only XylA part of the wild-type xylose promoter version XylWT (Bba_K1741007).
pxylS-E1_5'UTR has an E1 enhancer introduced into promoter???s 5???UTR. E1 is an RBS from gene 10 of bacteriophage T7. This promoter is induced by xylose (0.4%), which starts the transcription of the ORF.
Construct contains his-tagged sfGFP protein.

Notes

We started working on xylose promoters, because of the cheap inducer-xylose and potentially tight control of expression. Modification of E1-enhancer in 5'UTR gives the highest protein expression levels.

Xylose promoter's sequence comes from E.coli genome, E1 enhancer comes from gene 10 of bacteroophage T7.
