Description
Ycdo-Cas9 is a fusion of the Ycdo signaling sequence to the N-terminus of staphylococcus aureus CRISPR associated protein 9 (saCas9). Ycdo utilizes the sec pathway to direct the fused protein to the periplasm of the cell. Therefore, by fusing Cas9 to the C-terminus of the signaling sequence, it should be directed to the periplasm of the cell.
Notes
Ycdo was ordered as a gblock with homology to the BBa_K2019000-BBa_R0040 expression system. BBa_K2019000-BBa_R0040 was linearized at the 5' end of saCas9. A 2 piece gibson assembly was then performed with the gblock and Cas9 expression backbone.
