Description
This part contains the VioC gene codon optimized for yeast (BBa_K2165000) attached to the CUP1 inducible yeast promoter (BBa_K2165004) and the ADH1 terminator. The inclusion of an inducible promoter in this construct makes it easier to control the color output of the violacein pathway by altering the concentration of cupric ions in the cell environment to control the level of expression in the presence of other genes in the violacein pathway.
Notes
The VioC gene was codon optimized for use in yeast to optimize response time of the violacein system due to an introduction of cupric ions.
Source
VioC is derived from the violacein pathway in Chromobacterium Violaceum, a type of Proteobacteria. The CUP1 promoter was identified in the 5' untranslated region of the metallothionein gene in S. cerevisiae. The ADH1 terminator is also from the genomic DNA of S. cerevisiae.
