Description
This part contains the VioD gene codon optimized for yeast (BBa_K2165001) attached to the Gal1 inducible yeast promoter (BBa_J63006) and the ADH1 terminator. The inclusion of an inducible promoter in this construct makes it easier to control the color output of the violacein pathway by altering the concentration of galactose in the cell environment to control the level of expression in the presence of other genes in the violacein pathway.
Notes
The VioD gene was codon optimized for use in yeast to optimize response time of the violacein system due to an introduction of galactose.
Source
VioD is derived from the violacein pathway in Chromobacterium Violaceum, a type of Proteobacteria. The Gal1 promoter is from the genomic DNA of S. cerevisiae in the GAL1 locus. The ADH1 terminator is also from the genomic DNA of S. cerevisiae.
