Description
Construction of a genetic toggle switch in E. Coli. Collins Toggle, Advanced Biotechnology, BU.The Collins UV Toggle (2000) biobricked. It is a bistable toggle (on or off state) and switching is induced by UV exposure or IPTG. If the toggle is set with IPTG, the cells will express cI which will inhibit Plambda. If this is exposed to certain levels of UV (see power modulations) cI is cleaved by Rec-A (a UV induced enzyme) and lacI expression begins and inhibits Ptrc.Notes
We used PCR to biobrick this sequence from its original form in an Amp plasmid with SC101 origin. We had to transfer into pSB1C3 to submit it to the registry.Source
