Description
This device is based on gene encoding small RNA (sRNA), which represses ChiP gene synthesis by pairing with a sequence in 5??? untranslated region (UTR) near ribosome binding site (RBS) of chip messenger RNA (mRNA). (Caught at its own game: regulatory small RNA inactivated by an inducible transcript mimicking its target. Figueroa-Bossi et al. 2009, Genes and development 23:2004-2015) This leads to degradation of the targeted mRNA by enzymes called RNases. (Lauren S. Waters and Gisela Storz, Regulatory RNAs in Bacteria, Cell 136, 615-628, 2009) The repression occurs in the absence of the substrates (chitooligosaccharides) stimulating chip gene transcription.
