Notes

Both TetR and yeast codon optimized BFP were designed without a stop codon to enable the construction of a fusion protein with BFP and the rtTA activation domain.

Source

The LacI repressor was PCR amplified from a plasmid source of a biobrick (BBa_K319046) that was submitted last year by the uOttawa team. Yeast codon optimized BFP was synthesized by the uOttawa team using Mr. Gene DNA synthesis. The rtTA activation domain was PCR amplified from a plasmid obtained by our host lab.
