DescriptionThis actuator is designed to produce beta-galactosidase with an N-terminal secretion tag in HeLa cells. It links the alpha and omega fragments of the E. coli lacZ gene, though the codons have been optimized for expression in HeLa cells. It includes the kozak sequence immediately before the start codon and a secretion tag immediately after the start codon.
NotesThe lacZ alpha fragment and lacZ omega fragment were both codon optimized for expression in HeLa cells. After combining all the parts, there were only 7 repeats of ≥10 base pairs, so in order to maintain codon optimization, no further changes were made.
