Description
TetR-promoter driven chimeric receptor containing the N-terminal transmembrane sensor domain (1..442) of the quorum-sensing receptor LuxN from Vibrio harveyi fused to the C-terminal signaling domain (216..551) of the E.coli MCP receptor, Tsr. The domains are linked by an NdeI site (translation = 'HM').
Notes
The internal NdeI (CATATG) site of the Tsr C-terminal fragment 2 occuring at 772bp of the original protein receptor gene was mutated to (CACATG). This allowed the fusion of the LuxN and Tsr fragments by cloning in an NdeI site upstream at 646bp of Tsr, to test the effects of different fragment lengths.
