Description
This MammoBlock composite device produces the pDisplay-Vasopressin-MYC synthetic ligand designed to activate expression from the AVPR2-TEVs-Gal4-VP16 signaling receptor. Because this ligand is displayed in a membrane-bound, extracellular fashion, it is ideal for juxtacrine signaling that is orthogonal to other juxtacrine signaling systems, particularly the Notch-Delta system. Transcription is driven by the Hef1a-LacO promoter and therefore repressible by LacI variants.
Notes
In order to adhere to the recombination-based MammoBlock standard, and because shipping Biosafety Level 2 mammalian expression vectors to the Registry of Standard Biological Parts is not currently possible, we are providing the sequence resulting from Gateway recombination of the constituent MammoBlock parts here without redistributing the physical DNA plasmids to the Registry. In future years, we hope to make use of improvements in the Registry's accomodations to provide our complete expression vectors.
