Description
This MammoBlock composite device produces the Notch-Gal4-ESN juxtacrine signaling receptor when induced with Gal4 transactivator variants and otherwise produces Notch-Gal4-ESN at a low, basal (OFF) level. This device is applicable to cell patterning experiments employing the Notch-Delta system and is generally useful for the interrogation or engineering of multicellular phenomena that rely upon cell-cell contact. Note that in this particular case, activation of Notch-Gal4-ESN will release the same transactivator necessary for production of Notch-Gal4-ESN, creating a positive feedback loop upon activation.
Notes
In order to adhere to the recombination-based MammoBlock standard, and because shipping Biosafety Level 2 mammalian expression vectors to the Registry of Standard Biological Parts is not currently possible, we are providing the sequence resulting from Gateway recombination of the constituent MammoBlock parts here without redistributing the physical DNA plasmids to the Registry. In future years, we hope to make use of improvements in the Registry's accommodations to provide our complete expression vectors.
