Description

It is a device that contained a promoter (BBa_R0010), RBS (BBa_B0034) for each desaturase gene ∆9,12 and ∆15. When free fatty acids, for example, stearic acid (18:0) enters cell, it is converted into oleic acid (18:1∆9) by ∆9 desaturase, then to linolenic acid (18:2∆9,12) by ∆12 desaturase and finally to alpha-Linolenic acid, ALA (18:3∆9,12,15) by ∆15 desaturase. This device is designated to express under the presence of IPTG.

Notes

The size of the whole device could be relatively large. The selection of promoter is important as it controls when the machine starts producing desaturases. The investment of producing 3 enzymes in a time could be a energy burden for E. coli.

Source

The amino acid sequences of desaturases genes are derived from Synechocystis sp., but codon optimised for expression in E. coli.
