Description

Consist of six fragments from magnetosome island of Magnetospirillum gryphiswaldense genome.

Due to potential difficulty in introducing the large magnetosome formation operons into Azotobacter vinelandii, a "template" consists of flanking sequences from mamAB operonsexcept the mamO operon is designed for homologous recombination.

Part containing this template (K1648XXX) together with another part with mamO operon(K1648XXX) is introduced into A. vinelandii genome via random integration before full operon sequences to be transformed into A. vinelandii.

Notes

In order to form a homologous recombination template, we selected 250 bp of 5' and 3' ends from mamHIEJKLMN and mamPAQRBSTU (parts of mamAB operon without the mamO operon) as flanking sequences. Fragments are joined via overlapping PCR.
