Description
CrtEBI with RBS that is assembled by previous team is created by standard assembly of Part BBa_K118014 (CrtE with rbs), Part BBa_K118006 (CrtB with rbs) and Part BBa_K118005 (CrtI with rbs), which are submitted by previous iGEM teams. Together, enzymes CrtE, CrtB and CrtI convert colourless farnesyl pyrophosphate to red lycopene (via intermediates geranylgeranyl pyroiphosphate and phytoene). And then crtY(lycopene B-cyclase) converts lycopene to orange beta-carotene. The RNA thermometer that can be used for temperature sensitive post-transcriptional regulation which is based on a RNA thermometer from Salmonella Enterica Tyhpy initiates translation at 37??C. Therefore crtY expresses significantly at 37??C. The native terminator is included.
Notes
We design this composite biobrick to show red and orange pigment at different temperature. Lower than 37??C the pigment is red lycopene and at 37??C the pigment is orange.
